Materials and methods. Carbonized rice husk with nanostructured surface of carrier was used. Three from 12 strains including L.acidophilus, L.fermentum and L.plantarum with expressive antimutagen activity are used. Ames bacterial test system on strains S. typhimurim TA 98 and ТA 100 was used. The genoprotector properties of consortium are investigated by means of the algorithm including bacterial Rec- and SOS- bacterial systems and the metaphase analysis of a marrow of white mice. The mice model of antibiotic-induced dysbiosis caused by peroral application of ciprofloxacin and treatment effect of immobilized probiotic was used.

POLYCOMPONENT IMMOBILIZED PROBIOTIC WITH ANTIMUTAGEN ACTIVITY

The purpose. Construction the bacterial consortium with Lactobacillus strains immobilized on nanostructured carbonizing matrix.

Materials and methods. Carbonized rice husk with nanostructured surface of carrier was used. Three from 12 strains including L.acidophilus, L.fermentum and L.plantarum with expressive antimutagen activity are used. Ames bacterial test system on strains S. typhimurim TA 98 and ТA 100 was used.  The genoprotector properties of consortium are investigated by means of the algorithm including bacterial Rec- and SOS- bacterial systems and the metaphase analysis of a marrow of white mice. The mice model of antibiotic-induced dysbiosis caused by peroral application of ciprofloxacin and treatment effect of immobilized probiotic was used.

Results. Selections strains of lactobacilli, possessing high gen protections activity concerning modelling of mutagen. Completely chokes mutagens, induced N-nitrozo-N-metilurina and 2 nitrofluaren, estimated on frequency reversion in his- locus at S. typhimurim TA 100 and ТA 98. Mutagen effects, decreases on 97 %. Number of chromosomal aberrations in cages of a marrow of the mice, induced to straight lines mutagen N-nitroso-N-metilurina and promutagen N-nitrosomorpholins, at introduction in strains of lactobacilli food allowance in animals decreases on 70 % and 75 % accordingly. The decrease of genotoxic excrements at the experimental animals receiving immobilized lactobacilli after preliminary mutagen loading is revealed; in fecalase activity of procancerogenic β-glucuronidase enzyme decreases on 43 %. It is shown that new immobilized probiotic characterized by more expressed therapeutical effect at antibiotic-induced experimental dysbiosis.

Conclusion. It is developed a novel immobilized probiotic contained three Lactobacillus strains adsorbed on nanostructured carbonizing matrix with the expressed antimutagen and treatment action.
